H16. BALB/c for N-ethyl-N-nitrosourea(ENU) Mutagenesis

N-ethyl-N-nitrosourea(ENU) induces varied mutation as a powerful mutagen in mice. We injected intraperitoneally 2X100mg/kg ENU and 4X100mg/kg ENU into BALB/c male mice. After ENU injection, we investigated temporary sterile period, fertility and reproductive performance with breeding program specially designed. In the 2X100mg/kg group, the temporary sterile period was 15 weeks after ENU injection, and the recovery rate of fertility by 30 weeks after ENU injection was 69%. But in the 4X100mg/kg ENU group, the fertility did not recover by 30 weeks after ENU injection. In the 2X100mg/kg ENU group, the number of average pups were 2.1 per litter, and the control group, the number of average pups were 5.3 per litter. In the 2X100mg/kg ENU group, the weaning rate was 66% from the 17th week to the 35th week. We could not have reproductive in the 4X100mg/kg group, because there was no delivery by 35th week after ENU injection. All F1(43 heads) & F2(527 heads) mice were screened for morphological phenodeviants. We have obtained of 53 phenodeviant mice. Morphological malformation, physiological & behavioral abnormality were appeared such as small body size, rolling, bent tail, tail tip loss, short tail, small eye, toe short, finger syndactylia, closed eye and tail tip bleeding. It is supposed to discover the useful phenodeviants with ENU on the basis of basic data gotten at this study.
