Each year, the rate of incidence of obesity and obesity related diseases in the developed world continue to rise. Very few of these cases have so far been attributed to known monogenic causes, even though it is a highly heritable trait. Previously we have reported the discovery of four body fat quantitative trait loci (QTL), between the long-term divergently selected fat (F) and lean (L) -lines of mice, differing 5-fold in total body fat. We have since generated a congenic line for the Chr15 Fob3-QTL region by backcrossing to the F-line, and have carried out a more detailed QTL mapping analysis and microarray analysis between these two lines. Here we report the analysis of the differentially expressed transcripts in liver and brown adipose tissue found through our microarray study. Out of 15000 cDNAs screened we found 5 differentially expressed transcripts, and two of these mapped to the Chr15 Fob3-QTL region. Northern analysis was used firstly to confirm the microarray results for all 5 candidates and to perform a detailed developmental and tissue distribution analysis on the two Chr15 Fob3-QTL-associated transcripts.
