The clinical-chemical screen of the German Mouse Clinic identifies and characterises mouse mutants with defects of various organ systems or changes in metabolic pathways, asociated with aberrations of clinical chemical parameters or hematological parameters and blood coagulation to model human diseases like diabetes mellitus, anemia, coagulopathies, and others. The methods used are appropriate routine procedures, allowing the screening of large numbers of mice for a broad spectrum of parameters. In the primary clinical-chemical screen 19 different parameters are measured including various enzyme activities, as well as plasma concentrations of specific substrates and electrolytes. The high throughput of the screen is insured by the use of a Olympus AU 400 autoanalyzer. Additionally, we measure 8 basic hematological parameter with a blood analyzer (ABC-Animal Blood Counter, Scil), which has been carefully validated for the analysis of mouse blood. In a secondary screen specific profiles of organs are performed if certain parameters of the primary screen show significant deviations from the baseline levels. For example, an iron profile consists of the measurement of iron (Fe), ferritin (Fer), transferrin (Trf), unsaturated iron binding capacity (UIBC), and lactate dehydrogenase (LDH). Additionally, a broad spectrum of urin analyses using the AU 400 autoanalyzer, measurement of the coagulation time tested in whole blood to identify coagulation disorders, and the analysis of differential white blood counts by microscopic examination of blood smears are possible.
