DNAs were isolated from animal bones (1,200 - 1,300 years BP) excavated from Kwagji Cheju, Korea) archeological sites. The mitochondrial DNA (mtDNA) sequences were amplified from the DNA using polymerase chain reaction(PCR). The oliginucleotide primers used in this study are the highly conserved mtDNA region which specifies a 377 base pair (bP) fragment of the cytochrome b gene (primers L14841, H15149), and 571 bp fragment in the hypervariable region of bovine mtDNA. Comparing the sequences with those of Cheju native cattle and Cheju native swine, we can estimate the differences or similarities between the ancestral and existing animals.
