H14. Deletion Complexes in the Mid-To-Distal Region of Mouse Chromosome 15

Mice that carry chromosomal deletions are valuable reagents that can be used to facilitate genetic and functional mapping of the corresponding region. One of their uses is the fine-mapping of point mutations induced by ENU (N-ethyl-N-nitrosourea) to a defined position on the chromosome. An ongoing mutagenesis project at Oak Ridge National Laboratory is the recovery of recessive mutations induced by ENU in the mid-to-distal region of Chr 15 (~30 cM, homologous to human 8q, 22q, and 12q). We are interested in generating deletion complexes covering the distal half of Chr 15 for fine-mapping these recessive mutations and for serving as reagents to facilitate gene identification in the region. We employ the efficient strategy of inducing deletions by radiation in F1 hybrid ES cells to generate the deletion complexes. To induce deletions in the ES cells by radiation, neo-tk targeting vectors have been constructed for four deletion-focal-points (DFPs): Oc90 (37.5 cM), Sox10 (46.6 cM), Glycam1 (63 cM) and Pde1b1 (distal end). Additional DFPs will be selected to cover the entire region with deletion complexes if necessary. The current progress of this project will be reported.
