Within the framework of the German Human Genome Project (DHGP) we have implemented a platform for the functional analysis of mouse mutant lines from the Munich ENU mutagenesis screen using RNA expression profiling technology on a large scale and high throughput basis. The routine analysis of RNA expression patterns from organs will elucidate the molecular biology of such mouse mutants and provide new insights on mammalian gene function.
To achieve this goal the establishment of standardised protocols from mouse husbandry and tissue sampling to the molecular techniques of DNA-chip hybridisations was crucial. We have generated high quality DNA glass chips for genes with known functions and new genes with unknown functions derived from representational difference analysis (RDA). Software for the selection of gene specific ESTs (used as probes) and for the normalisation and visualisation of expression data has been developed. Our results show that RNA expression profiles of organs from different individuals are highly reproducible. Several candidate genes for differential gene expression in mouse mutant lines have been identified. Expression profiles that have been obtained in a first high throughput series are currently being used to establish a comprehensive database system for gene annotation.
