POSTER 41 - QTL MAPPING ANALYSIS OF ENDOCRINE REACTIVITY TO RESTRAINT STRESS IN BN AND WKHA RATS

The influence of genetic factors on stress reactivity has been largely demonstrated but the underlying molecular mechanisms remain undefined. In this study, we looked for QTL associated with endocrine responses to acute restraint stress in a F2 intercross progeny from BN and WKHA rats. These rat strains differ for their ACTH and corticosterone concentrations as well as for plasma renin activity in response to a 20 min restraint stress. QTL mapping analysis on the F2 progeny revealed a suggestive male specific QTL on chromosome 3 (named Pra3) and a significant QTL (LOD score=5,2)on chromosome 2 (named Pra2) both associated with renin activity. Phenotypic effects of the Pra2 and Pra3 alleles are opposed. Angiotensin II type 1b receptor (Agtr1b) maps in Pra2 confidence interval and constitutes a good positional candidate to explain part of the differential reactivity of the renin-angiotensin system after acute stress between BN and WKHA rats.
