We are using an exchangeable gene trap to isolate and characterize novel genes involved in mouse development. Here, we report the analysis of Ayu8008 mouse line obtained by using gene-trapping vector pU-Hachi.
The beta-geo was expressed in the yolk sac, somite and neural tube at E8.5 and E9.5 embryos. At E13.5 and E15.5, the beta-geo was expressed in the brain, heart and muscle. At adult, the expression was detected mainly in the brain, heart, lung, kidney, testis and skin.
The trapped gene was a novel gene and showed similarity to several ESTs. We detected two kinds of transcripts by RT-PCR and expected the transcripts were produced by alternative splicing.
The phenotype of the homozygotes was dependent on the genetic background. The chimeric mice, whose genetic background is mixed with C57BL/6 and CBA, were backcrossed to C57BL/6. The homozygotes (F1 x F1) were viable and appeared to be phenotypically normal. However, the homozygotes (F3 x F3) were showed apparent growth retardation and the homozygotes (F6 x F6) were not viable by 1 week old.
