Abstract ADAM family consists of a number of transmembrane proteins containing a disintegrin and metalloprotease domain. The ADAM proteins have both cell adhesion and protease activities and act in a highly diverse set of biological processes including fertilization, neurogenesis, myogenesis and inflammatory response. ADAM22 is highly expressed in the brain. The adam22-/- mice presented severe ataxia and died before weaning, but the function of ADAM22 is still unknown. In present work, the protein 14-3-3zeta interacting with ADAM22 was detected by yeast two-hybrid assay. 14-3-3 is widely involved in many signal transduction pathways related to important processes of living cells, but this is the first time to demonstrate that 14-3-3 can interact with an ADAM family member. The specificity of interaction between ADAM22 and 14-3-3zeta was proved by binding assay and immunoprecipitation. We had also localized the major binding site of 14-3-3zeta in the last 28 amino acid residues of ADAM22 cytoplasmic tail. The interaction between ADAM22 and 14-3-3zeta suggests that the ADAM22 may play a crucial role in neural function and development.
