Mouse mutants with behavioral phenotypes are powerful tools to elucidate a molecular basis of behavior. In order to establish a large number of mouse mutants, we have initiated dominant screening for behavioral mutations in Mouse ENU random mutagenesis program. Behavioral phenotypes we focus on are abnormal rest/activity cycles, excessive fear/anxiety, and learning/memory deficits, which are common symptoms of human neuropsychiatric disorders. For the first screening of these complex traits, we are conducting home-cage activity test (rest / activity cycles), open-field test (fear/ anxiety), and passive avoidance test (learning and memory). In home-cage activity test, a mouse is individually housed under a 24 hr light:dark cycle (LD 12:12), and spontaneous activity are monitored for 5 consecutive days. In open-field test, locomotor activity in a novel open field is measured for 20 min to detect a novelty-induced activity burst or behavioral suppression. In the passive avoidance test, the latency to entering a dark chamber is recorded as an index of learning after the delivery of an electrical shock in the dark chamber.Thus far, we have screened about 1,500 F1 progeny derived from ENU-injected C57BL/6J males, and more than 60 outliers have been detected. Most of them were found in open-field test. We have confirmed inheritance in several lines which show open-field high activity. Others are currently undergoing inheritance tests.
