LARGE SCALE GENE FUNCTION AND DRUG DISCOVERY IN THE POST-GENOME ERA

We have amassed a library of over 200,000 mouse embryonic stem cell clones using high-throughput gene trapping technologies that disrupt gene function and enable large-scale reverse genetics in the mammal. We have used this resource called OmniBank to create a relational database of trapped genes fueling our phenotypic analysis of 1000 genes per year covering all members of the ‘druggable’ gene families. Lexicon’s phenotyping protocol is designed to detect medically valuable targets for therapeutic intervention via small molecule, antibody, or therapeutic protein development.
Using defined preclinical endpoints, we have benchmarked our phenotyping protocol with relevant pharmaceutical agents to maximize our efficiency of novel target discovery. Our experience gives some indication of the significance of gene knockouts for pharmaceutical development and the number of drug targets the genome may contain.
