Phospholipid transfer protein (PLTP) is important in the transfer of phospholipids between lipoprotein particles and the remodeling of HDL. Previous studies suggest that plasma PLTP activity is regulated at the transcriptional as well as at the protein level. To identify loci that are involved in the regulation of PLTP activity, we performed quantitative trait locus (QTL) analyses on plasma PLTP activity using 217 male F2 progeny from an intercross between SM/J and NZB/B1NJ inbred mice fed either chow or an atherogenic diet. Two significant QTL for plasma PLTP activity on chow (Pltpq1 and Pltpq2) were found on chromosomes 3 (34 cM, LOD 3.5) and 10 (66 cM, LOD 4.1), respectively. Two other significant QTL for PLTP activity on the atherogenic diet (Pltpq3 and Pltpq4) were found on chromosomes 9 (56 cM, LOD 4.5) and 15 (34 cM, LOD 5.0), respectively. In addition we found one significant QTL (Plti1) for the inducibility of PLTP activity on chromosome 4 (70 cM, LOD 3.7). Several candidate genes were tested by sequence comparison and expression studies for these five QTL that could be involved in the regulation of PLTP activity both at the transcriptional (Nr1h4, Ppara, and Scap) and the protein level (Apoa1bp and Apof).
