F38 Positional Cloning of the Flexed-Tail Locus

The flexed-tail (f/f) mouse mutant shows defects in red blood cell development that leads to fetal-neonatal anemia and the inability of the adult mice to rapidly respond to erythropoietic stress. We are attempting to clone the f locus by positional mapping. We initially generated a genetic linkage map of the f locus using a (BALB/c x C57BL/6-f) F1 intercross. Radiation Hybrid mapping of this region using the T31 Mouse RH panel (Jackson Laboratory Mouse RH Map) demonstrated that the two flanking markers (D13Mit13 and D13Mit250) could not be separated. Subsequent physical mapping of the region showed that D13Mit13 and D13Mit250 were contained on a single BAC clone of 100Kb. Currently, we are exon trapping and characterizing transcription units in this region.
